hybcell is the world's first cylindrical microarray, whereas DNA oligos are immobilised on a cylindrical coated surface. Matching DNA fragments in the sample bind to these probes on the surface. A fluorescence readout shows these interactions.
The hybcell - and therefore the array itself - is constantly moved (rotated) through the sample. In this way, the sample is rapidly and homogeneously agitated towards the probes of the surface, resulting in improved reproducibility, drastic reduction in reaction times and flexible assay formats (e.g. a sequence of scans and heating and/or cooling steps.
Unless common hybridization of DNA-probes on the microarrays surface, compact sequencing continues with an enzymatic reaction on the spots (solid phase primer extension), followed by a stringent washing step. Intention of compact sequencing is the differentiation of target DNA down to a single base.
compact sequencing is a two-step process. First step is a simple, single-plex PCR reaction (e.g. 16S). This first step provides single-stranded fluorescence labelled DNA for the second step. The second step is the proprietary compact sequencing step carried out in the hyborg.
The whole process takes less than two hours and the sequencing step in the hyborg utilises a patented, extremely fast heating and cooling method.
The DNA sequence information is automatically generated by the software, identifying which of the microarray spots (oligos) have been elongated along the attached PCR products.
