# 16S & 28S amplicon sequencing

> GINA and PCR-Box also work outside the hybcell workflow: as sample preparation and PCR for your own 16S and 28S amplicon sequencing — Sanger or NGS.

### Amplicon sequencing · 16S / 28S, Sanger or NGS
**16S** pan-bacterial PCR · plus 28S pan-fungal

PCR-Box Bacteria is a pan-bacterial 16S PCR mix, PCR-Box Fungi a pan-fungal 28S PCR mix. Beside compact sequencing on the hybcell, the amplicons are ready for state-of-the-art 16S and 28S sequencing (Sanger, NGS). From whole blood, GINA first depletes human DNA and enriches the microbial DNA.

- Preparation: Enrichment of microbes
- Preparation: DNA extraction
- Preparation: Columns or magnetic beads

- Ready-to-use PCR mixes, single reactions in 0.2 mL tubes
- Extremely sensitive, with the possibility of RNA transcription
- Hot-start functionality and UNG carry-over protection
- Manufactured under cleanroom conditions

Workflow:

01. Sample preparation: GINA 500
02. DNA extraction & PCR: GINA DNA Purification, 3rd party DNA extraction, PCR-Box Bacteria, PCR-Box Fungi
03. Sequencing: Sanger (third-party), NGS (third-party)

## 16S sequencing — literature
The cube dx PCR has been compared with in-house PCR and 16S sequencing:
