hybcell is the world's first cylindrical microarray. Detector molecules, such as DNA oligos or antibodies are immobilised on coated surfaces. Certain compounds in the test sample bind to these molecules and a readout is made to detect this interaction. As opposed to carrying out the incubation and reaction between analytes and detector molecules in a standard two dimensional format, the patented hybcell technology is centred on two concentric cylinders, with detector molecules being linked to the outer surface of the inner cylinder. The test sample is pipetted into the hybcell through a central channel in the hybcell and the sample sinks to the bottom. The hybcell is sealed with a rubber-like lid and the rack of 8 hybcells is inserted into the hyborg. The hybcell is moved to the reaction area in the hyborg and the inner cylinder is rotated within the fixed outer cylinder. In this way, the reaction mixture is rapidly and homogeneously agitated between the inner active surface and the transparent outer surface. Key benefits of the hybcell are:
Improved reproducibility: The rotation of the inner cylinder causes shear forces which efficiently agitate the liquid and ensure a homogeneous reaction mixture. Like regular stirring when cooking, it ensures robust and reliable reactions across all parts of the microarray.
Drastic reduction in reaction times: As opposed to passive diffusion which is traditionally a limiting factor in flat 2D microarrays, the active diffusion caused by rotational mixing drastically reduces reaction to few minutes.
Flexible assay formats: Due to the unique design of the hybcell, all crucial processes like liquid handling, heating and scanning can be freely combined with only a few limitations. This flexibility enables advanced assay formats like compact sequencing and compact profiling.
Following the preparation of DNA template, Anagnostics compact seqeuncing is used to detect presence of either specific DNA sequences (e.g. corresponding to bacterial or fungal species, antibiotic resistance genes) or of specific and single base pair mutations (e.g. KRAS, BRAF, EGFR). Compact sequencing is a two-step process. Firstly, there is a straightforward PCR step, followed by a proprietary sequencing step carried out in the hyborg. The whole process takes less than two hours and the sequencing step in the hyborg utilises a patented extremely fast heating and cooling method. The DNA sequence information is generated by identifying which of the microarray spots have elongated PCR products attached to them by the software automatically.
To cover a wide dynamic range, compact profiling combines the concepts of competitive (for high concentrations) and sandwich (for low concentrations) formats in a single test. Starting from native sample (urine, saliva, whole blood, serum or plasma) the process is carried out in the hyborg and takes between 15 and 20 minutes. Quantitative results are derived by the software automatically and presented in comprehensive reports.
Both proprietary assay technologies have been successfully used in a number of CE-IVD certified tests and are therefore proven to be robust and reliable in the clinical setting.
hyborg operating software is a simple to use yet powerful tool to organise device runs, operate hyborg and analyze and archive results. All reports generated by the software deliver results without the need of interpretation of raw data (like e.g. melting curves or peaks of sequencing reactions). Therefore time to result is minimal, outcome does not depend on individuals and processes and results are electronically documented. Outstanding is the software's ability to present trends of results over time for a single patient.
